Human placental extract rescues hippocampal damage associated with cognitive impairment in diabetic male rats through antioxidative, anti-inflammatory, and neuromodulatory activities

记忆和认知障碍已成为与糖尿病相关的重要共病,但有效的治疗方法仍然难以找到。多项研究表明,人胎盘提取物(HPE)富含具有抗炎、抗氧化、抗凋亡和免疫调节作用的生物活性分子。然而,HPE对记忆和认知衰退的影响尚不明确。本研究旨在调查HPE对链脲佐菌素(STZ)诱导的大鼠记忆和认知功能障碍的治疗效果,并探讨其潜在机制。为了诱导2型糖尿病(T2DM),先给大鼠喂食高脂饮食两周,然后单次腹腔注射STZ(40 mg/kg体重),随后用HPE(20 mg/kg体重/天)治疗14天。行为测试结果显示,在接受STZ处理的大鼠中,HPE显著增强了学习、记忆和认知功能。具体而言,HPE在新物体识别测试中的辨别指数从STZ大鼠的负值增加到治疗大鼠的正值,并改善了T迷宫中的自发交替行为,从41.25 ± 8.25%提高至74.50 ± 8.50%。此外,HPE还显著提高了血清中胰岛素、血糖水平、胰岛素抵抗的稳态模型评估(HOMA-IR)以及脂质谱与未治疗糖尿病大鼠相比,并调节了海马组织中的氧化应激标志物、抗氧化剂、促炎细胞因子和神经化学物质水平,突出了其抗氧化和抗炎特性。值得注意的是,HPE治疗改善了海马的神经形态学并减少了DNA损伤。带有尾巴的细胞百分比从糖尿病大鼠的25.67 ± 0.33下降到接受HPE治疗后的13.67 ± 0.88。总之,HPE具有神经保护作用,并可能作为针对T2DM相关神经退行性症状的有前景的治疗方法。
