STAMBPL1/TRIM21 Balances AXL Stability Impacting Mesenchymal Phenotype and Immune Response in KIRC

肾透明细胞癌(KIRC)被认为是一种具有免疫原性的肿瘤,几十年来免疫疗法已成为其治疗的一部分。通过上皮-间充质转化(EMT),获得肿瘤间质表型与免疫逃逸相关,并可能导致对免疫疗法的抵抗。本文报告了STAM结合蛋白样1(STAMBPL1)在KIRC细胞中发展间质和免疫逃逸表型中的作用。从机制上看,STAMBPL1通过减少TRIM21介导的AXL蛋白K63连接泛素化并随后将其降解于溶酶体中,从而增加了AXL蛋白丰度及其表面积累,进而增强了间质基因表达的同时抑制了趋化因子CXCL9/10和HLA/B/C。此外,STAMBPL1通过促进p65核转位而增强PD-L1转录,并且敲低(KD)STAMBPL1可增加PD-1阻断的抗肿瘤效果。此外,沉默STAMBPL1与酪氨酸激酶抑制剂(TKI)索拉非尼在抑制KIRC方面也表现出协同作用。总体而言,靶向STAMBPL1/TRIM21/AXL轴可以减少间质表型并增强癌症治疗的抗肿瘤疗效。
