<enabstracts>Introduction: Plant stoichiometric traits can indicate nutrient allocation and resource-use strategies, but their interpretation depends on organ identity and developmental stage. Fargesia denudata is a staple bamboo for giant pandas and an important clonal understory plant in subalpine forests. We examined how C, N, and P concentrations and their ratios are coordinated across aboveground organs and ontogeny, and how stage-specific stoichiometric economics is associated with environmental and community variables. Methods: Across 52 plots in Wanglang National Nature Reserve, including 37 plots with F. denudata, we measured stoichiometric traits in shoot culms and in mature culms, leaves, and twigs, together with topographic, soil, and community variables. Results: Stoichiometric coordination was modular and stage-specific, with limited independent within-organ coordination retained only in shoot culms and mature twigs, and cross-organ links suggesting that mature twigs acted as the main bridge between culms and leaves. PC1 axes generally separated N- and P-enriched stoichiometry from ratio-enriched stoichiometry, indicating comparable but developmentally reorganized stoichiometric-economic axes. Shoot culms occupied the nutrient-enriched, relatively acquisitive end of the axis, whereas mature culms shifted toward the ratio-enriched, more conservative end. Shoot-culm PC1 did not scale significantly into mature organ-level or integrated mature aboveground PC1, indicating developmental reorganization rather than direct continuity. In mature bamboo, aboveground stoichiometric economics was assembled through organ-specific contributions, with twigs forming the main link between culms and leaves. Plots with F. denudata occurred on steeper slopes and had higher soil C concentration than plots without F. denudata. Environmental and community associations were stage-dependent and were concentrated mainly in shoot culms. Elevation showed a significant positive total effect on shoot-culm PC1, corresponding to a shift toward a more conservative stoichiometric strategy along the elevational gradient. Mature aboveground PC1 showed a more limited pattern, with only a single correlation with soil pH. Discussion: These results suggest that stoichiometric strategies in F. denudata are shaped by organ-specific coordination, ontogenetic reorganization, and stage-dependent links with environmental and community conditions. Organ- and stage-specific stoichiometric indicators may inform bamboo resource assessment and giant panda habitat evaluation. <enabstracts>The ecological stoichiometric traits of forest floor material nutrient resorption in forest ecosystems. However, systematic insights into nutrient allocation and scaling during the transformation of plant organs to forest floor material remain limited. This study examined six representative tree species on the southern slope of the Qilian Mountains, quantifying the carbon (C), nitrogen (N), and phosphorus (P) contents and their stoichiometric characteristics in leaves, branches, and mixed forest floor material. Allometric relationships between N and P were analyzed using the standardized major axis regression. Coniferous species exhibited a conservative strategy with high C/N and C/P ratios, whereas broad-leaved species and mixed forests exhibited nutrient-enrichment strategies. During the organ-to-forest floor material transformation, N and P contents significantly reduced, whereas C/N and C/P ratios increased, indicating strong nutrient resorption. N and P were positively associated with plant organs and forest floor material, with isometric relationships (b = 1.06 and 0.98 for plant organs and forest floor material, respectively). Because the slopes did not differ significantly from 1 (p > 0.05), the N:P ratio remained relatively constant, with no significant P limitation. This indicates that tree species regulate forest floor material quality through divergent nutrient utilization strategies, modulating nutrient cycling. This study provides a theoretical foundation for the ecological restoration and stand structure optimization of alpine forests.
Doublesex gene influences sex differentiation and embryonic development in predatory mite Phytoseiulus persimilis

<enabstracts>Background: Phytoseiulus persimilis is an effective biocontrol agent characterized by paternal genome elimination (PGE), an unusual reproductive system in which males eliminate the paternal genome during embryogenesis. However, the molecular mechanism underlying sex determination and reproductive regulation in this species remain poorly understood. Results: Transcriptome-based analyses identified two doublesex (dsx) homologs, Ppdsx1 and Ppdsx2, as candidate regulators of reproduction. Weighted gene co-expression network analysis (WGCNA) assigned Ppdsx2 to a pre-mating-associated co-expression module enriched for reproductive and signaling pathways. Functional analyses revealed clear divergence between the two genes. RNA interference (RNAi) of Ppdsx1 reduced the proportion of female offspring, whereas RNAi of Ppdsx2 induced sex reversal, developmental abnormalities, and impaired egg viability. Yeast two-hybrid and glutathione S-transferase (GST) pull-down assays further demonstrated interactions between Dsx proteins and vitellogenin (Vg)-derived fragments identified from a complementary DNA (cDNA) library screen, suggesting a previously unrecognized connection between sex determination and reproductive nutrient allocation. Conclusions: Ppdsx1 contributes to maintenance of the female developmental pathway, whereas Ppdsx2 represents a strong candidate component of the PGE-associated sex-determination cascade. The observed Dsx-Vg fragment interaction suggests a potential link between reproductive developmental programs and nutrient allocation pathways. These findings provide new insights into the molecular basis of sex determination and reproductive regulation in phytoseiid mites and establish a foundation for future studies on the coupling of reproductive development and resource allocation. © 2026 Society of Chemical Industry. <enabstracts>The restoration of soils contaminated by antibiotic resistance genes (ARGs) is essential for agricultural safety and human health. Although biochar and silicon (Si) fertilizer have been commonly applied individually as amendments for soil remediation and rice quality improvement, their combined effects remain unclear. Here, we systematically assessed the effects of the integrated application of biochar and Si fertilizer on rice nutrient uptake, soil properties, bacterial community structure and functions, and ARG abundance by conducting a 120-day pot experiment with four treatments: control (CK), 0.15 t ha-1 of Si fertilizer (SF), 3 t ha-1 of biochar (BC), and their combination (SC). The results showed that the combined treatment enhanced rice panicle length and promoted the translocation of Si and potassium to leaves, while altering the distribution of nitrogen and phosphorus in rice. In soil, co-application did not markedly alter pH, SOM, or most available nutrient levels, but significantly increased TN and TP contents. Moreover, the SC treatment resulted in the lowest abundances of detected ARGs and MGEs among all treatments. It also reshaped the bacterial community composition and functional profiles, including reduced abundance or functional contribution of key r-strategist genera associated with ARGs and nitrogen/phosphorus metabolism, such as Dinghuibacter and Roseomonas. Overall, the co-application of biochar and Si fertilizer improved rice nutrient supply, enhanced soil nutrient status, and mitigated the dissemination of ARGs. These findings provide a scientific basis for sustainable paddy field production and ARG risk management in paddy ecosystems. <enabstracts>The basal endosperm transfer layer (BETL) constitutes the indispensable gateway mediating maternal nutrient transport into developing maize kernels, a process that fundamentally determines grain-filling efficiency and final grain yield. To date, only a few transcription factors have been documented to directly regulate BETL formation and nutrient transport, and the transcriptional cascades governing BETL cell differentiation remain largely elusive. Here, we functionally characterized ZmHB118, an HD-Zip IV transcription factor with exclusive expression in the BETL. Loss of function of ZmHB118 produced smaller kernels, accompanied by impaired cell wall ingrowth (CWI) in the BETL and reduced grain-filling rate. Integrative RNA-seq and DAP-seq analyses identified two BETL-specific, ZmHB118 target genes, encoding non-canonical lipid transfer proteins. Indeed, ZmHB118 robustly transactivates these two downstream genes, named NCLTP1 and NCLTP2, suggesting its role in mediating lipid transport toward the cell periphery to maintain proper CWI formation. Overall, our findings establish ZmHB118 as a master transcriptional regulator of BETL formation and maternal-to-filial nutrient allocation during grain filling, thus providing a promising genetic target for and new insights for boosting maize grain yield.
A Wild Emmer Wheat B3 Transcription Factor Regulates Seed Growth and Nutrient Allocation

<enabstracts>Seed weight (SW) and nutrient allocation are key determinants of yield and grain quality in wheat, yet the regulatory basis of naturally occurring variation in these traits remains poorly resolved. Wild emmer wheat (Triticum dicoccoides), the progenitor of modern wheat, retains extensive eco-geographically structured genetic diversity that was largely eroded during domestication. Here, we identify a B3-domain transcription factor (B3TF) as a key regulator of seed growth and metabolic partitioning in wheat. Genome-wide association analysis of ~460 wild emmer accessions reveals a major locus on chromosome 2BL associated with SW, seed area and nitrogen (N) content, displaying pronounced climatic differentiation across environmental gradients. Introgression of the 2BL wild segment into the hexaploid wheat cultivars Chinese Spring and Bethlehem increases SW in cultivated backgrounds. Independent loss-of-function alleles generated by EMS mutagenesis in the tetraploid wheat cultivar Kronos produce larger seeds. Further, RNA-seq of EMS mutants revealed metabolic reprogramming with upregulated fatty acid, nitrogen and phenylpropanoid pathways and downregulated carbohydrate metabolism and sugar transport. Metabolomic, lipidomic and ICP-MS data showed increased essential amino acids, sugars, lipids, N content and minerals (Zn, Fe, Mo). Furthermore, CRISPR/Cas9-mediated editing in the hexaploid wheat cultivar Fielder produced similar increases in SW and N content as observed in the EMS mutants, establishing this gene as a negative regulator of seed growth across ploidy levels. In addition, natural haplotypes show reciprocal climatic distributions, linking regulatory variation to environmental adaptation. Our findings uncover a TF underlying natural seed trait variation in wild wheat, providing a framework for exploiting regulatory alleles to enhance yield and nutritional quality in modern wheat. <enabstracts>Nitrogen (N), phosphorus (P), and potassium (K) are essential elements for plants and are also the main limiting elements in the growth process. In order to investigate the effects of fertilization on the nutrient content and stoichiometric ratio of Pinus yunnanensis seedlings, we implemented a complete 3 × 3 factorial design incorporating three nitrogen levels (0, 0.4, and 0.8 g·plant-1) and three phosphorus levels (0, 3, and 6 g·plant-1), yielding nine distinct treatment combinations. The contents of N, P, and K in different organs of P. yunnanensis were determined, and their stoichiometric ratios were calculated. N content ranked as needle > root > stem; P content was in the order of root > stem > needle; and K content showed the pattern of stem > needle > root. Fertilization did not alter the above nutrient distribution patterns; it significantly influenced nutrient concentrations and stoichiometry. Single applications of nitrogen fertilizer had the most significant effect on nutrient content; compared with the unfertilized control, single nitrogen application increased needle N content by 1.13-fold. Combined nitrogen and phosphorus application preferentially promoted the accumulation of N relative to P in stems; specifically, under combined N-P treatment, stem N content increased by 1.11-fold while stem P content increased by 1.03-fold, indicating a greater relative increase in N. In terms of stoichiometric ratio, fertilization increased N:P, particularly in the root and needle; root N:P increased by 1.19-fold under T8 (0.8 g N + 3 g P) and needle N:P increased by 1.17-fold under T2 (3 g P alone), both indicating a mitigation of N limitation. Consistent with the nutrient content results, single nitrogen application exerted the greatest effect on stoichiometric ratio, followed by nitrogen and phosphorus combined application, and single phosphorus application had the least effect. These findings suggest that fertilization can alter nutrient allocation and stoichiometric relationships in Pinus yunnanensis, which may affect its growth and metabolic processes, but further studies are needed to link these changes to seedling growth. <enabstracts>In alpine conifer forests, nutrient acquisition is shaped by nitrogen (N), phosphorus (P) and cold soils, but the elevational adjustment of nutrient-acquisition traits in Abies fabri is still poorly known. Along the Gongga Mountain transect (2,800-3,400 m), soil inorganic N declined markedly with elevation, whereas available phosphorus, defined here as Bray-1 extractable P, did not differ significantly among elevations. Forests at 2,800, 3,100 and 3,400 m were sampled for soil nutrients, leaf N:P, foliar N and P fractions, root morphology, root exudation, mycorrhizal colonisation, root enzyme activities and non-structural carbohydrates (NSC) to test how leaf nutrient status was linked to belowground acquisition traits. The trait pattern did not follow a directional available-P gradient. With elevation, foliar N allocation shifted from Rubisco towards cell-wall pools, while foliar P shifted towards metabolic and nucleic-acid pools. Fine roots showed a more conservative morphology, with lower specific root length (SRL) and specific root area (SRA), whereas fine-root acid phosphatase (APase) and N-assimilating enzyme activities increased. With the high-elevation decline in leaf N:P, A. fabri responses reflected N-P stoichiometric context, internal nutrient allocation and root-trait coordination rather than a simple soil-P availability gradient.
Interspecific interactions alter survival strategies of tree-shrub plants and hinder treeline ascent at alpine treelines in Southeastern Tibet: Evidence from13C and15N

<enabstracts>Nitrogen (N) and phosphorus (P) are pivotal nutrients that support species diversity in plant communities. However, in nutrient-poor alpine treeline ecosystems, the mechanisms through which interspecific interactions among treeline plants regulate nitrogen uptake patterns, competitive capacity, and survival strategies across functional groups remain poorly understood. Using four plant communities in the alpine treeline of Sygera Mountain (southeastern Tibet) as study subjects, we employed in situ 15N labeling to quantify inorganic nitrogen uptake rates in the leaves, branches, and fine roots of the dominant species in these communities. We further clarified the survival strategies, directionality of interspecific interactions, and competitive strength of two key treeline plant species under conditions shaped primarily by interspecific interactions and mediated by natural environmental factors. The results revealed three key findings: (1) Water use efficiency (WUE) was higher in mixed communities than in monospecific communities. Both Juniperus saltuaria and Rhododendron nivale preferred nitrate nitrogen (NO3--N), and this preference increased in mixed communities for both species, with the increase in R. nivale substantially greater than that in J. saltuaria. (2) Within mixed communities, the inorganic nitrogen uptake rates of leaves, branches, and fine roots in R. nivale were significantly higher than those in J. saltuaria. Interspecific interactions exerted a facilitative effect on R. nivale but an inhibitory effect on J. saltuaria, and soil total phosphorus (TP) was identified as the key nutrient supporting their growth and survival. (3) Compared with pure J. saltuaria stands, J. saltuaria-dominated mixed communities allocated a higher proportion of nutrients to aboveground organs and therefore adopted an acquisitive nutrient strategy. In contrast, relative to pure R. nivale communities, mixed communities dominated by R. nivale preferentially translocated nutrients to root systems, reflecting a defensive and conservative nutrient allocation pattern. Overall, this study confirms that the interspecific relationships and nutrient strategies of J. saltuaria and R. nivale in the Sygera Mountain alpine treeline are consistent with the Competitive Exclusion Hypothesis, Facilitation Hypothesis, and Productivity-Nutrient Allocation Hypothesis. From physiological and ecological perspectives, this study elucidates the regulatory mechanisms shaping interspecific relationships among alpine treeline plants. The findings advance our understanding of how biological factors, particularly interspecific interactions, modulate treeline migration under climate warming and provide critical theoretical support for endangered species protection and ecological restoration in this high-altitude region.
Pumpkin rootstock-driven variation in mineral nutrient allocation determines differential melon fruit quality

<enabstracts>Rootstocks are commonly used in melon cultivation to mitigate biotic and abiotic stresses and improve fruit quality. However, the extent to which rootstocks influence nutrient allocation, growth dynamics, and fruit quality formation in the scion remains insufficiently understood. This study compared the effects of two pumpkin rootstocks (Tianzhen No. 1 and Sizhuang No. 12) on melon fruit quality by examining nutrient partitioning, sugar accumulation, hormonal balance, and expression of sugar metabolism-related genes. Compared with non-grafted plants, rootstock grafting enhanced plant growth and increased total N, P, and K uptake (mg plant-1) as well as tissue nutrient concentrations (mg g-1 DW) during vine growth and fruit maturation stages. Tianzhen No. 1 showed higher K distribution to the scion (97.6%) and greater fruit K accumulation (45.88 mg/g) than Sizhuang No. 12. Fruits grafted onto Tianzhen No. 1 also exhibited higher total soluble solids (15.96 °Brix) and soluble sugar contents, including glucose (18.28 mg g-1), fructose (20.20 mg g-1), and sucrose (144.25 mg g-1), whereas fruits grafted onto Sizhuang No. 12 showed higher firmness (4.18 kg cm-2) compared with Tianzhen No. 1-grafted fruits (3.07 kg cm-2). At fruit maturation, Tianzhen No. 1-grafted melons had higher ABA content (14.49 ng/g) and upregulated expression of sugar transporter genes (HT2, HT7, SWEET4, SWEET7) and genes encoding key metabolic enzymes (SPS1, NIN3) compared to Sizhuang No. 12. These observed associations suggest that rootstock-mediated differences in nutrient uptake may contribute to altered scion fruit quality metabolism. Overall, the findings provide insights into the physiological and molecular basis of rootstock-dependent variation in melon fruit quality and may contribute to the development of targeted rootstock selection strategies in melon breeding programs.
